Aprotinin's inhibitory activity against these proteases makes it a valuable tool in various biochemical, biotechnological, and medical applications.

A competitive and reversible inhibitor of esterase and protease activity. Forms a tight complex with and blocks the active site of target enzymes. Inhibits a number of different proteases, including chymotrypsin, coagulation factors involved in the pre-phase of blood clotting, kallikrein (Kd = 1 X 10-7 M), plasmin (Kd = 2.3 X 10-10 M), tissue and leukocyte proteinases, and trypsin (Kd = 5 X 10-14 M). Does not inhibit Factor Xa or thrombin. Reported to be relatively acid and heat stable. Effectively inhibits target proteases at equimolar concentration. Useful as a serine proteinase inhibitor during purification of proteins and in studies of zymogen activation systems. Reported to suppress virus HA cleavage and limit reproduction of human and avian influenza viruses with a single arginine in the HA cleavage site.

General Description Trypsin inhibitor, pancreas type from bovine lung. It is known as Pancreatic trypsin inhibitor (BPTI). Aprotinin, also known as pancreatic trypsin inhibitor and trypsin-kallikrein inhibitor, is found to be expressed in lungs, spleen, liver, and pancreas. It is also found to be present in the free form in calf serum.
Biochem/physiol Actions Aprotinin is a competitive serine protease inhibitor that forms stable complexes with and blocks the active sites of enzyme. This binding is reversible, and most aprotinin-protease complexes will dissociate at extreme pH levels >10 or <3. Structurally, Aprotinin is a monomeric globular protein derived from bovine lung that consists of 58 amino acids, arranged in a single polypeptide chain with three crosslinking disulfide bridges.
