General description

Trypan Blue (C.l. 23850) is a large, tetrasulfonated anionic, hydrophilic dye. It is also known as diamine blue 3B.

Application

Trypan blue has been used as a dye in trypan blue exclusion assay/cell viability assay to detect dead cells.

Biochem/physiol Actions

Trypan Blue is excluded by most living cells, but can be taken into phagocytes and certain other cells.

This dye is useful to differentiate the dead and live cells based on the cell′s ability to take up or exclude the dye. A viable cell is assumed to possess an intact cell membrane and hence excludes this dye and the cytoplasm appears clear. While on the other hand non-viable cells shows a blue cytoplasm.
Trypan blue is routinely used for viability testing for diverse cell types including frozen sperm and aortic muscle cells exposed to antifungal agents. It is also employed as a collagen stain in a Van Gieson procedure. To stain amyloid, Trypan blue is considered as a blue alternative to Congo red in Puchtler-Bennhold stains. It is suitable for use in cell viability assay:
  • to determine the number of living and necrotic cells by staining with 0.2% trypan blue solution.
  • to stain plant tissue in order to visualize cell death.

Uses Trypan Blue is an azo based, hydrophilic, tetrasulfonated blue acid dye widely utilized for assessing cell viability. As an easy method to determining the viability of cells, Trypan Blue will stain dead cells with permeable membranes blue, while the dye is excluded by most living cells and their intact membranes thereby allowing visual determination of living versus dead cells. Although Trypan Blue is predominately used for assessing cell viability of cultured cells, other applications for this dye have been reported. In rat abdominal organ slices Trypan Blue has been utilized to determine cell viability as well as islet cell cluster cell viability from caprine pancreatic samples. Dyes and metabolites.

Reactivity Profile Direct Blue 14 is an azo compound. Azo, diazo, azido compounds can detonate. This applies in particular to organic azides that have been sensitized by the addition of metal salts or strong acids. Toxic gases are formed by mixing materials of this class with acids, aldehydes, amides, carbamates, cyanides, inorganic fluorides, halogenated organics, isocyanates, ketones, metals, nitrides, peroxides, phenols, epoxides, acyl halides, and strong oxidizing or reducing agents. Flammable gases are formed by mixing materials in this group with alkali metals. Explosive combination can occur with strong oxidizing agents, metal salts, peroxides, and sulfides.
